PURPOSE:To obtain a new subject compound having tachykinin antagonism and effective in treatment and prevention for tachykinin-intercalated diseases such as asthma, atopic dermatitis and nettle rash, by culturing a specific microorganism belonging to the genus Aspergillus. CONSTITUTION:A microorganism belonging to the genus Aspergillus and having ability capable of producing a compound (TAN-1666A) expressed by formula I or a compound (TAN-1666B) expressed by formula II [e.g. Aspergillus terreus FL-29694 (FERM P-13126), etc.] is inoculated into a culture medium and subjected to shake culture at 28 deg.C for 48hr to afford a seed culture. The seed culture is transplanted to a culture medium in a stainless-steel tank and subjected to spinner culture under aeration at 24 deg.C and then, pH of culture medium is adjusted to pH 2-4 and the cultured mixture is extracted with dichloromethane, etc., and the extracted liquid is extracted with a dilute alkali water. Thereby, TAN-1666B is dissolved in a water layer and TAN-1666A is left in an organic solvent layer and these layers are purified to provide the objective TAN-1666 related compound expressed by formula III [R1 is H; R2 is (substituted)alkoxy, etc., or R2 forms direct bond together with R1]. |
