PURPOSE:To diagnose diseases, etc., by cleaving a nucleic acid sample and a standard nucleic acid sample with a restriction enzyme, binding a labeled linker to both terminals, forming both samples in a state of single strand, subjecting the both samples to gel electrophoresis and comparing both patterns and judging variation of the nucleic acid sample. CONSTITUTION:A nucleic acid sample 2 containing a variant part 3 and a standard nucleic acid sample 1 for comparing with the nucleic acid sample 2 are prepared and these samples are proliferated by PCR method to afford gene copies 5-2 and 5-1 and each subjected to restriction enzyme treatment to cleave the gene copies at a restriction enzyme-recognizing sites 6. A labeled linker 11 obtained by labelling one side chain of double stranded chain DNA oligomer with a labelling material is bound to both terminals of these cleaved each nucleic acid fragments 7-2 and 7-1 to afford a nucleic acid sample fragment group 12-2 having labeled both terminals and a standard nucleic acid sample fragment group 12-1 having labeled both terminals and these fragments are formed into single stranded states and subjected to gel electrophoresis and variation 3 of the nucleic acid sample 2 is judged by comparison of the resultant electrophoresis pattern to diagnose diseases, etc., with the gene. |
