PURPOSE:To obtain a simple and highly sensitive method for detecting DNA or RNA. CONSTITUTION:A fluorescent labeled RNA probe 2 having a sequence complementary to a single stranded target DNA 1 and having a terminal to which fluorescent label 10 is bound is mixed with a specimen and the mixture is hybridized with a target DNA 1. Then, ribonuclease H is added thereto. A double stranded RNA of DNA-RNA hybrid 3 is selectively decomposed by the ribonuclease H. The target DNA 1 returned to single strand is again hybridized with RNA probe 2 and the RNA probe 2 is similarly decomposed into RNA probe small fragments, whose numbers are amplified, by Ribonuclease H and these fragments are isolated. This reaction process is automatically repeated. Then, the reaction is stopped and the reaction liquid is subjected to separation by electrophoresis and the RNA probe decomposed into small fragments is detected. Thereby, detection of target DNA can readily be carried out. |
