PURPOSE: To provide the medicine for thrombolysis, etc., which is obtd. by vaccum drying an unbuffered and refrigerated aq. soln. of tissue plasminogen activation factors of a specific pH range, provides an effect only at the point where clot exists and has no danger of hemorrhage. CONSTITUTION: The tissue palsminogen activator (t-PA) having the amino acid sequence expressed by the formula or having the same amino acid sequence except that the 245 - the amino acid from the serine N-terminal is valine in place of methionine or lacking any of the first three amino acids in some ceases or having the additional polypeptide N-terminal precedent sequence added with the formula Gly-Ala-Arg in some cases are dissolved into an aq. sodium chloride soln. contg. a surfactant to prepare the unbuffered aq. soln. adjusted to pH 3.5 and, thereafter, this soln. is diluted with a mannitol soln. and is subjected to freezing at -35 deg.C, then to vacuum drying. The medicinal compsn. which provides the effect only at the place where the clot of a fibrin network structure exists and has no danger of the hemorrhage is thereby obtd. |
