PURPOSE:To provide the subject new derivative obtained by depleting or substituting a specific amino acid of interleukin-1beta, having high activity to suppress the proliferation of tumor cell, activity to promote the production of cytokinin and anti-inflammatory action and useful as an antitumor agent, an immune stimulation agent, an anti-inflammatory agent, etc. CONSTITUTION:cDNA is synthesized by using mRNA separated from human peripheral blood by conventional method and a cDNA library is prepared from the cDNA. The library is screened to select a DNA coding interleukin-1beta and the DNA is subjected to restriction enzyme treatment to obtain a DNA coding a polypeptide having the amino acid sequence of formula I wherein one or more amino acid residues of the 1st Ala, the 3rd Val, the 4th, 11th and 98th Arg, the 5th and 153rd Ser, the 8th and 71st Cys, the 30th His, the 93rd, 97th and 103rd Lys, the 99th Phe, the 120th Trp and the 121st Tyr are deleted or substituted, one or more amino acid residues of the 1st to 9th and 103rd to 153rd residues are deleted and an amino acid sequence of formula II, etc., is added to the N-terminal. The objective peptide derivative is produced by the expression of the DNA. |
