PURPOSE:To provide the subject medicine containing a substance such as a lactoferrin- hydrolyzed peptide having a specific amino acid sequence as an active ingredient, having a brain-protecting activity, useful for preventing ischemic cerebral diseases, etc., reduced in adverse reactions and good in its heat resistance and antigerminal property. CONSTITUTION:The brain-protecting agent comprising a peptide of the formula (Xaa is the residue of an arbitrary amino acid excluding Cys), etc., its derivative and salt, etc., or their mixture comprising two or more of the compounds is obtained by dissolving bovine lactoferrin in purified water, adjusting the pH of the solution to 2.5 with a 0.1N hydrochloric acid, hydrolyzing the bovine lactoferrin in the solution by the addition of swine pepsin at 37K!n for 6 hr, adjusting the hydrolyzed solution to a pH of 7.0 with a 0.1N sodium hydroxide aqueous solution, heating the solution at 80 deg.C for 10min for the inactivation of the enzyme, cooling the product, centrifuging the cooled solution, collecting the transparent supernatant, subjecting the supernatant to a high performance liquid chromatography using a 20% acetonitrile containing O.05% trifluoroacetic acid and subsequently using a 20-60% acetonitrile gradient containing 0.05% trifluoroacetic acid, collecting the eluted fractions and subsequently vacuum-drying the collected products. |
