PURPOSE:To provide a new cDNA probe useful for the diagnosis of nephropathy, the treatment of nephropathy, etc. CONSTITUTION:The objective probe is a clc-klcDNA probe characteristic to the RNA expressing a chloride channel locally existing in the ascending limb of Henle's loop of kidney and complementary to the RNA and has for example the base sequence of formula. It can be prepared by preparing a single-stranded cDNA by the reverse transcription of the RNA of the mammal kidney, subjecting the cDNA to PCR using 5'-CCGGATCCCTNGGNGA(A/G)GA (T/C) TGGAT(T/C/A)TT(C/T)CT-3' (sense chain) and 5'-CCGAATTC(A/G/T)AT(A/G) TGNAC(A/G)AANGGNCC(T/C)TC(T/C)TT-3' (antisense chain) as degenerate primers and screening the renal cDNA library of mammal by using the PCR product as a probe. |
