摘要 Combination of haematopoietic growth factors contains interleukin(IL)-1,3- and -6. erythropoietin (EPO) and stem-cell growth factor (SCF). Also new are solns. contg. this combination. Pref. the soln. contains 10-100 (pref. 50-150) ng/ml IL-1; 1-1000 (pref. 50-150) U/ml IL-3, 10-1000 (pref. 50-150) U/ml IL-6; 0.1-10 (pref. 0.5-1) U/ml EPO and 10-1000 (pref. 50-150) ng/ml SCF. Opt. it also contains gamma interferon (INFG) at 1-1000 (pref. 50-150) U/ml. Peripheral blood precursor cells are isolated, pref. by immunoaffinity adsorption or fluorescence-activated cell sorting, to enrich CD34 positive cells. These are then cultured in presence of a soln. of the new combination, in autologous plasma or in nutrient medium, which may be serum free or contain animal serum. USE/ADVANTAGE - The compsn. is used for ex vivo propagation of haematopoietic precursor cells, specifically CD34 positive cells. The propagated cells are partic. used after high dose chemotherapy for rapid reconstitution of haematopoiesis. The compsn. generates precursor cells with high clonogenic potential and a high proportion of cells able to form multiline colonies (i.e. terminal differentiation is inhibited). Partic. omission of colony-stimulating factors (present in known compsns.) increases the no. of clonogenic cells, while addn. of IFNg further increases the no. of nucleated and colony-forming cells. Not only are myeloid precursors amplified but also early haematopoietic and erythrocyte precursors. Large no. of precursors can now be generated from a small amt. of blood, avoiding the need for multiple apheresis stages.
