PURPOSE:To measure an alpha-amylase isoenzyme in a reaction process of one stage without the influences of intrinsic glucose, etc., by using coloring reagents such as multo-oligosaccharide represented by a specific formula. CONSTITUTION:An electrophoresis film sheet having a specimen containing an alpha-amylase isoenzyme thereto is subject to electrophoresis operation so as to fruction it. The obtained electrophoresis film sheet is pregnated with a coloring reagent containing a malgo-oligosaccharide, etc., represented by a formula I [where, A: group shown by a formula II (R1, R2, X1, and X2 stand for hydrogen atom, lower alkyl group, oxygen atom, sulfur atom, respectively); F: fructose; G: glucose; n; integer of 3 to 15] and a compound that generates water-insoluble pigments through reduction. Then, after it is incubated, the colored pattern formed on the sheet is subject to colorimetric measurement. Thus, the alpha-amylase isoenzyme can be fractioned in a reaction process of one stage without any influence of intrinsic glucose, multose, etc. |
