<p>PURPOSE:To provide a new compound useful for medicines having in-vivo calcium-regulating activity and oncocyte differentiation-inducive activity. CONSTITUTION:The compound of formula I (R<1> is Hand R<2> is OH, or vice versa; X is H, OH or OH protected with a protective group therefor; R<3> is H or protective group for OH), e.g. 26,26,26,27,27,27-hexafluoro-1alpha,22s,25- trihydroxyvitamin D3. The compound of the formula I can be obtained by coupling reaction between (A) a ketone of formula II (R<4> is OR<6> and R<5> is H, or vice versa; R<6> is protective group for OH) and an anion derived from a phosphine oxide of formula III (R<7> is protective group for OH; Y is H or OR<7>; Ph is phenyl) followed by, where appropriate, deprotection. The coupling reaction is made in an inert atmosphere (e.g. argon) in an ether solvent (e.g. THF) at -100 to -50 deg.C for 10min to 24hr.</p> |
