Improved procedure for the preparation of 6-α- aminopenicillins, particularly ampicillin or amoxicillin, involves: (a) reacting an N-substituted vinyl α- amino acid with an acylating agent, within an aromatic solvent, preferably toluene, with which a mixed anhydride is obtained; and (b) reacting said mixed anhydride with a salt of 6-aminopenicillanic acid (6-APA) in an organic solvent, with which 6-α-aminopenicillins are obtained with a yield higher than 85% and a purity higher than 98%.
