摘要 |
PURPOSE:To produce the subject amino acid in high efficiency by reacting indole with serine in the presence of E.co1i containing a plasmid having fragments of genes governing the biosynthesis of tryptophan synthase, a proliferation controlling and distributing system and an autonomous replication capability. CONSTITUTION:A strain of E. coli is transformed by introducing a recombinant plasmid containing a DNA fragment containing a gene governing the biosynthesis of tryptophan synthase and originated from tryptophan operon, a DNA fragment containing a gene governing a proliferation controlling and distributing system and originated from F factor plasmid and a DNA fragment containing a gene governing the autonomous replication capability of ColE1 plasmid. The obtained transformed strain Escherichia coli K-12 YK2004 (FERM P-7838) is cultured in a medium and indole is made to react with DL-serine in the presence of the cultured product or its treated product to obtain L-tryptophan on an industrial scale in high efficiency. |
