PURPOSE:To obtain the title high-molecular weight elastin having excellent affinity for collagen molecule by treating pure insoluble elastin with pepsin under a specific condition to solubilize the elastin, then deactivating the pepsin and then removing low-molecular seed. CONSTITUTION:Spherical protein and lipid are removed from a raw material containing elastin, which is treated by an autoclave to remove collagen and further freeze-dried to give insoluble elastin. Then the insoluble elastic is treated with pepsin within 15 hours in an acidic solution at pH<=4 at 10-40 deg.C while stirring and solubilized. Then, the solubilized elastin is adjusted to pH12, pepsin is deactivated and further a low-molecular seed having 5,000-7,000 molecular weight is removed to give the aimed elastin having 640,000-8,300 molecular weight and >=1.5 as much molar ratio of acidic amino acid as that of insoluble elastin. |
