PURPOSE:To provide a new enzyme capable of decarboxylating pyrrole-2- carboxylic acid to form pyrrole and producing pyrrole-2-carboxylic acid from pyrrole in the presence of HCO3<->. CONSTITUTION:The enzyme having the following properties and its production process. (1) Action and substrate specificity, (a) decarboxylates pyrrole-2- carboxylic acid to form pyrrole, (b) produces pyrrole-2-carboxylic acid by using pyrrole as a substrate in the presence of HCO3<->: (2) optimum pH, 6.5 at 30 deg.C: (3) stable pH, 6.0-9.0: (4) optimum temperature range, optimum temperature is 45 deg.C: (5) thermal stability, stable up to 45 deg.C when maintained at pH6.5 for 10min: (6) molecular weight, subunit molecular weight is 50,000 measured by SDS electrophoresis and molecular weight is 98,000 measured by TSK-Gel G300OSW gel-filtration column: (7) Michaelis constant Km for pyrrole-2- carboxylic acid, 23.8X10<-3>M. |
