PURPOSE:To efficiently obtain the beer having a high fermentation degree by use of an inexpensive immobilized glucosidase in a state reduced in the leakage of the glucosidase by reacting the glucosidase with a cellulosic material subjected to a periodate oxidation treatment, and subsequently bringing a barley juice fermentation solution into contact with the covalently bonded immobilized enzyme. CONSTITUTION:A cellulosic material such as gauze is immersed in a 1% sodium periodate aqueous solution, shaken over a night at 4 deg.C for the periodate oxidation treatment of the cellulose, washed with distilled water, washed with acetone, dried, immersed in the solution of a glucosidase such as glucoamylase, left over two nights at 4 deg.C to covalently bond the glucosidase to the cellulosic material, and subsequently reduce the covalently bonded product with NaBH4 to produce an immobilized enzyme in which the bond is stabilized. The immobilized enzyme is immersed in a barley juice fermentation solution and subjected to a fermentation at a fermentation temperature of 10.5 deg.C for a month with stirring, thus providing the objective beer having a high fermentation degree with the inexpensive immobilized glucosidase without leaking the enzyme. |
