PURPOSE:To provide the subject protein having an antitumor activity and useful for the therapy of animal epithelial cancer, etc., by extracting the fruit body of Tricholoma matsutake with water, and subsequently successively subjecting the filtrate to a gel filtration column treatment, an anion exchange resin treatment, and a hydrophobic chromatography treatment for the isolation and purification of the protein. CONSTITUTION:The fruit body of Tricholoma matsutake is extracted with water, and the filtrate is subjected to a mol.wt. fractionation treatment with a gel filtration column to purify the protein. The protein is adsorbed to an anion exchange resin, eluted from the column with an eluting agent, further adsorbed onto a resin for hydrophobic chromatography, and again eluted from the column with an eluting agent to provide the pure protein having a mol.wt. of 100000-110000 as its sub unit by a SDS electrophoresis method and a mol.wt. of 200000-210000 by a gel filtration method, exhibiting a pale yellowish brown color to a pale brown color, and having an amino acid composition (molar ratio) of Ser:34.7, Arg:20.1, Asp:15.9, Phe:11.2, Ala:6.3, Thr:2.7, Tyr:2.1, Val:1.8, Ile:1.0, and Lys:0.7, Gly,Cys:0.5. |
