PURPOSE: To provide a method to separate and purify protein or polypeptide or to remove metal ions from a biological material and to provide a chromatography agent usable in this method and excellent in separation of substances having high affinity. CONSTITUTION: The chromatography agent having peculiar affinity is prepared by bonding phosvitin or modified phosvitin to Sepharose (R) matrix activated by cyanogen bromide. The obtained agent is used for separating and purifying protein or polypeptide, or removing metal ions from the biological material.
