A method for purifying at least one amphiphilic peptide or protein, wherein a material containing the at least one amphiphilic peptide or protein is contacted with a mixture of an aprotic organic solvent and an alcohol is added to form a single liquid phase, to which is subsequently added an aqueous material, such as a salt solution, to form a primarily aqueous phase containing the at least one amphiphilic peptide or protein, and an immiscible organic phase. The at least one amphiphilic peptide or protein in then recovered from the aqueous phase. In one embodiment, prior to contacting said material with a mixture of an aprotic organic solvent and an alcohol, a material containing at least one amphiphilic peptide or protein is contacted with a solution including an aprotic organic solvent, at an acidic pH, to provide an extract solution containing the at least one amphiphilic peptide or protein, and at least a portion of the aprotic organic solvent is removed from the extract solution to provide a remainder material containing the at least one amphiphilic peptide or protein. This method also may be used in multiple successive applications so as to purify further the at least one amphiphilic peptide or protein. These methods enable one to purify biologically active peptides or proteins without the use of costly purification agents and techniques.
|
