PURPOSE:To obtain the subject preparation useful for treating hypoproteinosis free from virus contamination, having excellent long-term stability by blending human serum albumin prepared by gene manipulation with acetyltryptophan (salt) and sodium caprylate. CONSTITUTION:A plasmid fragment having a transcription unit to manifest human serum albumin(HSA) is linked to a gene such as Pichia pastolis GTS11, which is inoculated into a medium, cultured at 30 deg.C for 3 days. The culture solution is centrifuged to separate the cells. The cultured supernatant liquid is collected, treated with a ultrafilter having 100,000-500,000 fractionated molecular weight, the filtrate is adjusted to proper pH, treated with a cation exchanger, a carrier for hydrophobic chromatography and an anion exchanger and purified to give HSA by gene manipulation. Then the aqueous solution of SHA is blended with acetyltryptophan or its salt and sodium caprylate as stabilizer to give the objective HSA-containing preparation stably preservable for a long period of time, free from the danger of contamination with hepatitis virus, etc., at all. |
