PURPOSE:To obtain phospholipase A1 capable of selectively hydrolyzing 1-position acyl group of a phospholipid by culturing a mold fungus belonging to Aspergillus in a culture medium and purifying the resultant product by column chromatography, etc. CONSTITUTION:A mold fungus belonging to Aspergillus is inoculated on a solid culture medium prepared by adding water to wheat bran and steaming the resultant mixture and subjected to koji-type culture at a prescribed temperature for a prescribed time. After completion of culture, a prescribed amount of water or a suitable buffer solution is added to koji and the resultant mixture is stirred and then subjected to pressure filtration to obtain an enzyme solution. Further, the obtained enzyme solution is purified by salting-out method, organic solvent precipitation method, gas chromatography column adsorption method, etc., to obtain phospholipase A11. The obtained phospholipase A1 exhibits an activity for producing a 2-acyl type lysophospholipid by selectively hydrolyzing only 1-position acyl group of a phospholipid. Reaction optimum pH is 3.5 to 4.5 and stable pH range is 5.5 to 10.5. Molecular weight is 37000 and an excellent activity and an excellent selectivity are exhibited. |
