<p>PURPOSE:To provide the monoclonal antibody capable of specifically recognizing a cancer-related mucin type sugar chain of cell surface, the structure of the sugar chain having been changed by the canceration of the cells, and useful for diagnosing and treating the cancer, etc. CONSTITUTION:The objective monoclonal antibody recognizing a glucolipid derivative of formula II and useful for the serum diagnosis, therapy, etc., of cancers is obtained by the steps of combining a sugar chain of formula I, which is estimated to be a kind of cancer-related mucin type sugar chain, to ceramide by a conventional method; allowing died Salmonella bacteria to adsorb the produced glucolipid derivative of formula II (Cer is ceramide); administering the adsorbed died bacteria into the abdominal cavity of a BALB/c mouse for its immunization; collecting the splenic cells of the mouse after subjected to the final secondary immunization; fusing the splenic cells with mouse myeloma cells in the presence of polyethylene glycol as a fusing agent; screening the antibody-producing strains by a solid phase enzymatic immune test; cloning the screened strain by a limiting dilution method; transplanting the obtained hybridoma into a mouse; recovering the antibody produced in the abdominal dropsy of the mouse; and subsequently purifying the antibody.</p> |
