<p>PURPOSE:To provide the monoclonal antibody specific to fats and oils, and to provide the hybridoma secreting the same. CONSTITUTION:A mouse, rabbit, etc., is immunized with the mixture of a carrier with an immunogen consisting of a triacylglycerol or diacylglycerol which is the ester of a fatty acid with glycerol, and antibody-producing cells are separated from the immunized animal. The antibody-producing cells are fused with myeloma cells, and their hybridomas are selected with a selection medium (e.g. a HAT medium). The presence or absence of the produced antibody in the supernatant of a hybridoma culture solution is detected in the presence of the immunogen and a serum by an ELISA method. After a hybridoma secreting the monoclonal antibody is separated, the hybridoma is subcultured in a medium The supernatant of the culture solution is subjected to a salting-out treatment and subsequently to a purification treatment by chromatography to recover the objective monoclonal antibody.</p> |
