<p>PURPOSE:To provide the monoclonal antibody capable of rapidly, highly sensitively and specifically detecting injurious bacteria in beers, beverages, foods, etc., by immunizing a mammalian with a lactic acid bacterium belonging to the third group of the genus Lactobacillus. CONSTITUTION:A lactic acid bacterium belonging to the third group of the genus Lactobacillus, such as Lactobacillus.brevis, Lactobacillus.collinoides, or Lactobacillus.suebicus, and separated from beer or a beer brewery is mixed with a Freund's incomplete adjuvant, and subsequently injected into the abdominal cavity of a BALB/c mouse for its immunization. After the mouse is finally immunized, the splenic cells of the mouse is collected, fused with mouse myeloma cells in the presence of 50% polyethylene glycol, and subsequently selectively cultured in a HAT medium to produce their hybridomas. The hybridomas are screened, and the obtained antibody- producing strain is cloned by a limiting dilution method, inoculated in the abdominal cavity of a mouse, and multiplied therein. After two weeks, the abdominal dropsy of the mouse is recovered, and the antibody is separated from the abdominal dropsy and then purified to provid the monoclonal antibody peculiar to the lactic acid bacteria belonging to the third group.</p> |
