PURPOSE:To provide a new cellulase having specific properties, useful for the enzymatic saccharification of cellulosic substances and the enzymatic treatment of paper pulp in a paper pulp-producing process, and having an effect to accelerate the fiber-disaggregating activity. CONSTITUTION:The objective cellulase A has following properties; e.g. (A) action: (i) the production of cellotriose, cellobiose, and glucose from an insoluble cellooligosaccharide, (ii) the production of water-soluble saccharides and water- insoluble saccharides from reduced phosphoric acid-swollen cellulose, and (iii)the acceleration of the fiber-disaggregating activity; (B) substrate specificity: exhibits a strong activity to CM-cellulose, and hydrolyzes the insoluble cellooligosaccharides, the phosphoric acid-swollen cellulose, cellotetraose, and cellopentaose; (C) optimal pH, pH stability: the optimal pH is 5, and the cellulase A is stable at a pH of 2-5, when CM-cellulose is used as a substrate; (D) optimal temperature, thermal stability; the optimal temperature is approximately 70 deg.C, and stable up to 60 deg.C, when the CM-cellulose is used as the substrate; (E) mol.wt.: approximately 53000 by a SDS polyacrylamide gel...slab phoresis method. The cellulase is obtained by culturing Trichoderma SP.SK-1919. |
