BIOTIN-INTRODUCING REAGENT AND METHOD FOR PURIFYING SYNTHETIC PEPTIDE WITH THE SAME

PURPOSE:To provide the subject reagent enabling to selectively separate only the objective mature peptide from the products of a solid phase peptide synthesis by the formation of an adipic acid-biotin bond to purify the synthetic peptide in a short time in a good yield. CONSTITUTION:The objective reagent comprises a biotin-introducing reagent used for selectively separating the mature peptide from a mixture system comprising the objective end amino group-containing mature peptide obtained by a solid phase peptide synthetic method and other end-capped non-mature peptides, and this reagent has a reactive terminal group at one molecular end and a biotinyl group at the other molecular end, and the reactive terminal group can form a bond which can be cleaved from the amino end of the mature peptide under a specific condition but which is stable for a protecting group- removing reaction under an acidic condition in the peptide-synthesizing process. A new compound of the formula, etc., are preferable as the reagent. The compound of the formula is obtained e.g. by reacting 2-[(N-biotinyl) aminoethylsulfonyl]ethanol with p-nitrophenylchloroformate.
