PURPOSE:To obtain a new protein, derived from a biological tissue and cell, capable of enhancing the production of an anti-DNA antibody and binding to the DNA and useful for understanding the morbid state, diagnosing and treating autoimmune diseases caused by abnormal production, transudation, etc., of nucleic acids such as the DNA. CONSTITUTION:A KML1-7 cell derived from a lymphoid established cell established as a cell strain capable of producing and releasing a factor enhancing the production of an anti-DNA antibody from the whole cell population of a tumescent lymph node in an MRL/l mouse is cultured in the presence of blood serum until a confluent state is attained. The cultured KML1-7 cell is then collected by centrifugation, transferred to a serum-free culture medium, suspended and subsequently cultured in a CO2 incubator for 3 days. The resultant culture solution is centrifuged to collect the culture supernatant and a serine protease inhibitor is directly added thereto. The prepared mixture is then concentrated with an ultrafiltration membrane and the resultant concentrate is adsorbed on an anion exchange resin. Common salt at >=0.2M concentration is added to purify the eluted fraction by high-performance liquid chromatography to afford the objective protein capable of enhancing the production of the anti-DNA antibody and binding to the DNA. |
