PURPOSE:To provide a new DNA useful for mass production of DSDGK of antiallergic pentapeptide by gene recombinant technique. CONSTITUTION:The DNA containing a base sequence coding the macromer peptide of antiallergic pentapeptide having the amino acid sequence of DSDGK, having e.g. a base sequence of the formula. With a base sequence coding a peptide as recurring unit, single-stranded DNA containing at least two such recurring units is mixed with another single-stranded DNA complementary thereto followed by thermal insulation and cooling to produce such a double- stranded DNA that both the two single-stranded DNAs are mutually complementary and slipped by at least one recurring unit, and dATP, dCTP, dGTP and dTTP are made to react in the presence of DNA polymerase to synthesize the DNA strand complementary to the single-stranded DNA region. The resulting double-stranded DNA is heated and dissociated into a single-stranded DNA and another single-stranded DNA complementary thereto. The operations ranging from the above-mentioned thermal insulation, cooling to the dissociation is repeated at least once, performing the operations of the thermal insulation, cooling and complementary strand synthesis. |
