GENE FOR ENHANCING TRANSFORMATION, USING MERCURY RESISTANT SHUTTLE VECTOR PLASMID FOR THE GENUS THIOBACILLUS

PURPOSE:To obtain a DNA factor and a host cell capable of transforming a cloning vector having mercury resistant selection marker for T. ferrooxidans in high frequency and stabilizing the vector in a state of vector plasmid CONSTITUTION:A connected DNA mixture containing a genome DNA of T. ferrooxidans E-15 strain and EcoRI fragment of Escherichia coli plasmid pUC 18 is subjected to cloning to a competent cell of Escherichia coli DH5alpha strain containing a chimeplasmid pAT116 to provide shuttle vector plasmid pTMR25 shown in the figure.
