Quick and simple recovery of ganglioside G LAC 2 from milk cream - by extn. with alkanol, concn. of anionic exchanger and chromatographic purificn., eliminating pollution of anionic solvents

Recovery of ganglioside Glac2 (I; (New Acoalpha)23Galbeta4Glcbeta1Cer) from milk cream comprises: (1) extracting the cream with a water-miscible extractant; (2) removal of the pptd. solid extract residue and reextn. of it with the same extractant; (3) purificn. of ganglioside from the extracts on an anion exchanger, and (4) chromatographic final purificn. of gangliosides. Pref. cream from bovine milk is used and the extractant is MeOh or EtOH, used at 2 vol. per vol. of cream. Step (4) is done on a silica gel column. Pref. insolubles in step (2) are removed by low-speed centrifugation or filtration. They are then extracted at least once more and gangliosides retained on the anion exchanger, then eluted in fractions. USE/ADVANTAGE - (I) is involved in tumour-associated phenomena and cell-cell recognition processes, and is used in research and industry (no details). Compared with known methods (which start from milk or buttermilk) this process is simpler, quicker, more economical and less polluting, partic. freeze-drying of the starting material is not necessary and expensive organic solvents are not required for 2-phase extn. Since fats remain in the pptn. during extn., the method is selective for gangliosides.
