<p>Method and kit for the preparation of nucleic acid, for detection and/or for amplification of said nucleic acid. The nucleic acid may be prepared by contacting a sample of cells, particularly whole blood, with an appropriate lysing agent under conditions in which red blood cells are lysed but white blood cells are not. The treated sample may then be centrifuged so as to separate desired white blood cells from non-desired red blood cells, and then purifying and lysing said desired white blood cells. The nucleic acid associated with the lysed cells can be detected and/or amplified, and the nucleic acid can also be screened for a certain nucleic acid sequence, such as a viral nucleic acid sequence. In particular, hemolysis may be performed using either CTAB or saponin, as illustrated in the figure.</p> |
