A method for detecting the presence or absence of more than one variant nucleotide in a target base sequence comprised in a nucleic acid sample using diagnostic primers having a terminal nucleotide complementary to either a suspected variant nucleotide or the corresponding normal nucleotide in the target base sequence and wherein the extension products of more than one diagnostic primer comprise a complementary overlap. Primers and kits for use in the claimed method.
