PURPOSE:To provide a new enzyme useful for producing valuable physiological activity-bearing sugar chains and modified products thereof, improving sugar chains bound to valuable physiologically active proteins, curing inflammation and metastases etc., and diagnosing diseases. CONSTITUTION:The alpha 3 sialyltransferase, which can be obtained by culture in a medium of the host cells (pref. 'namaluba' cells as a kind of human cells) transformed by a recombinant vector incorporated with cDNA coding this enzyme to produce and accumulate the aimed enzyme in the resulting cultured product. The above recombinant vector, which is also a new substance, can be obtained by the following process: a cDNA synthesized using, as template, mRNA extracted from animal cells is incorporated into a manifestation cloning vector and the resulting cDNA library is introduced into cells, and the resultant cells are cultured in the presence of lectin, and the cDNA coding the above enzyme is taken from the cells proliferated, and this cDNA is then introduced into the downstream portion of a promoter in the vector. |
