METHOD FOR OBTAINING RIBOSOMAL RNA RIBONUCLEASE WITH AN ACTIVITY INHIBITING THE PROTEINIC BIOSYNTHESIS FROM SEEDS OF CUCUMIS SATIVUS L., AND UTILIZATION THEREOF

It has been possible to obtain from the seeds of the plant Cucumis sativus L. a protein which behaves like a catalytic inhibitor of the biosynthesis of proteins both of procaryotic and eucaryotic systems. The protein that we have named cusativin has been obtained and purified to electrophoretic homogeneity through the following steps: a) obtaining saline extracts from seeds; b) obtaining acidifyed extracts from the prior extracts; c) chromatography through S Sepharose Fast Flow (Pharmacia-LKB); d) ultrafiltration through membrane YM-10 of Amicon; e) chromatography through Sephadex G 75 (Pharmacia-LKB); f) chromatography through CM Sepharose Fast Flow; g) ultracentrifugation through membrane YM-10 of Amicon; h) chromatography through Superdex 75-F.P.L.C. (Pharmacia-LKB); i) dialysis; j) chromatography through Mono-S (F.P.L.C., Pharmacia-LKB); k) dialysis and lyophilization of the resultant protein.
