PURPOSE:To enable mass production of a yellow pigment which is useful as a food additive because it is soluble in water, resistant to heat and light, stable in alkaline conditions and high in safety by culturing a mold strain in Epicoccum in a medium and separating and recovering the pigment accumulated in the medium. CONSTITUTION:A mold strain of Epicoccum, for example, Epicoccum nigrum sp-RDM-Y (FERM No. 12893) is inoculated with a platinum spatula to an enriched medium containing yeast essence and subjected to shaking culture at 25 deg.C for 7 to 10 days. Then, the culture liquid is centrifuged to separate into the supernatant and the cell bodies, the cell bodies are extracted with methanol and the extract and the supernatant are combined and subjected to ultrafiltration to remove high-molecular-weight components. Then, the filtrate is subjected to reverse osmosis to remove salts and low-molecular-weight components, the concentrate is passed through a adsorption resin column and the column is washed. Then, the adsorbate is eluted with 0.01% sodium hydroxide aqueous solution and the eluate is adjusted in its pH to 2.5 to 3.0 with hydrochloric acid to precipitate the yellow pigment, which is collected, dried to give the final product. |
