<p>PURPOSE:To obtain a glycerin derivative which made it possible to introduce a long-chain amino linker having arbitrary chain numbers into an arbitrary position in an oligomer such as DNA or RNA. CONSTITUTION:A compound of formula I [R<1> and R<2> are OH protecting groups or H; R<3> and R<4> are NH2 protecting groups, H, biotinyl or nonisotopic label or mutually bonded to constitute a ring; n is integer of >=5], e.g. 1,3- dibenzyl-2-(N-phenoxyacetyl-5-aminopentyl)-glycerin of formula II. The exemplified compound of formula II is obtained by reacting 1,3-dibenzylglycerin with a 5-halovaleronitrile to afford 1,3-dibenzyl-2-(4-cyanobutyl)-glycerin of formula III, reducing the compound of formula III and reacting the reduced compound with phenoxyacetyl chloride.</p> |
