PURPOSE:To simply and accurately identify lactic acid bacteria without using a reference sample by cleaving a ribosomal RNA gene of lactic acid bacteria with a restriction enzyme, analyzing length of the resultant DNA fragment and comparing the analyzed length with the length of a standard DNA. CONSTITUTION:A ribosomal RNA gene of lactic acid is cloven with a specific restriction enzyme HinfI, DraI, etc. Then, the length of the produced DNA fragment is analyzed and compared with a standard DNA strain to identify strain of lactic acid bacteria. Concretely, e.g. 16Sr DNA is specifically amplified from a chromosomal DNA by an enzymatic gene amplifying method and cloven with a restriction enzyme and then subjected to agarose gel electrophoresis and a test bacterium exhibiting electrophoresis pattern which coincides with a standard strain is identified as same bacterial strain as the standard strain. |
