An antibiotic effective against pleuro-pneumonia-like organisms in poultry, amethobottromycin, has the following properties: m.pt. 154-163‹ C.; is soluble in alcohol, esters, ethers, chlorinated solvents and benzene; is sparingly soluble in water and insoluble in hexane and petroleum ether; [α]<SP>20</SP>D= -25‹ in 95% ethanol; infra-red as in the drawing; contains C=58À40%, H=7À44%, N=12À42%, S=4À77% and 0=15À97%; possibly C 40 H 60 N 8 O 7 S; R f 0À30 in caprylalcohol phosphate buffer of pH 6À0; acid hydrolysis gives substances having a positive reaction with ninhydrin especially proline; forms salts with acids, e.g. HCl, tartaric and salicylic acids. Amethobottromycin is produced by cultivating Streptomyces canadensis ATCC 17776, or an amethobottromycin-producing mutant thereof, in an aqueous nutrient medium containing assimilable sources of carbon and nitrogen. The antibiotic is recovered from the fermentation broth by acidifying the broth to a pH of 4À8 to 5À8, filtering it, adsorbing the resulting filtrate on a strongly acidic cation-exchange resin, eluting the filtrate from the resin with aqueous methanolic ammonia, neutralizing the filtrate and extracting it with chloroform, performing partition chromatography on the chloroform extracts using chloroform and acetone as eluant, removing the solvent, dissolving the residue in methanol, acidifying to precipitate the crude antibiotic. The crude amethobottromycin is dissolved in 0À1 M phosphate buffer of pH 6À0 and subjected to partition chromatography on diatomaceous earth impregnated with capryl alcohol as the stationary phase, the amethobottromycin being contained in the fractions having an R f value of 0À30. Amethobottromycin, or a salt thereof, may be administered to poultry infected with pleuropneumonia-lik organisms, either orally or subcutaneously. |
