PURPOSE:To provide a new enzyme highly stable to surfactants and highly excellent as an enzyme for formulation in detergents. CONSTITUTION:The objective enzyme, alkali protease K-16N, having the following characteristics: (1) activity: hydrolyzing simple proteins or multiple proteins into oligopeptides or amino acids; (2) substrate specificity: giving favorable degradative activity on water-soluble proteins such as casein and hemoglobin and water-insoluble proteins such as keratin and elastin; (3) optimal pH: 10-11.5 when allowed to react in a buffer solution at 40 deg. for 10min using casein as substrate; (4) optimal temperature: 60 deg.C when allowed to react at pH10 using casein as substrate; (5) molecular weight: 15000 (gel filtration method), 14500+ or -500 or 12000+ or -500 (sodium dodecylsulfate-polyacrylamide gel electrophoresis); (6) effect of metallic Ca<2+>. This enzyme can be obtained through its production by Bacillus microorganisms [e.g. Bacillus sp. KSM-K16 (FERM P-3376)]. |
