Prodn. of monoclonal antibodies to human interferon alpha-2 - by hybridoma method with selection by ELISA assay, used in medical diagnostics, therapy monitoring and research, etc.

Prodn. of monoclonal antibodies aainst human interferon alpha-2) (I) is effected by immunising Balb/c mice with (I); fusing the mouse spleen cells with moude myeloma cells; cloning the resulting hybridomas in cell culture vessels; selecting antibody-producing clones by ELISA assay; and producing antibodies from clones that give extinction values at least 5 times greater than the blank value in the ELISA assay. The ELISA assay is pref. performed by binding (I) to microtitre plate wells, adding culture supernatant from the hybridoma clones, and detecting antibody binding by means of enzyme-labelled anti-mouse Ig. USE - The antibodies are useful in medical diagnostics, therapy monitoring, basic research and biotechnology. They are esp. useful for quantitative determn. of (I) and for affinity purificn. of natural or recombinant (I).
