PURPOSE:To enable the artificial culture of a large amount of Lyophyllum decastes (a kind of champignon) which has been unable to be proliferated by artificial culture by converting the mycelium of Lyophyllum decastes to protoplast with an enzyme, culturing the protoplast and selectively separating a strain capable of forming fruit body, thereby shortening the culture period. CONSTITUTION:Mycelium of Lyophyllum decastes is cultured on an agar medium at 25 deg.C for 2 weeks, inoculated to a liquid medium, cultured at 25 deg.C for 3 weeks under periodic shaking to thoroughly disperse the mycelia and separated by centrifugal separation. The collected mycelia are subjected to enzymatic treatment with e.g. cellulase, chitinase and glucanase to obtain a protoplast. The protoplast is suspended in a buffer solution and the suspension is smeared to a plate agar medium and cultured at 25 deg.C for 4 weeks. A strain capable of forming fruit body is selected and subjected to seed mycelium culture. The obtained seed mycelia are inoculated to an artificial culture base and cultured to enable the artificial culture of a large amount of Lyophyllum decastes which has been hardly culturable by conventional artificial culture method. |
