<p>PURPOSE:To obtain new protein chondromodulin-III useful as a therapeutic agent for fracture and various cartilage diseases, isolated from bovine fetal cartilage, having specific molecular weight, showing multiplication activity of cartilage cell and differentiation function promotion activity. CONSTITUTION:Ground bovine fetal cartilage is homogenized in a buffer solution by using polytron, etc., the homogenate is extracted at 4 deg.C for 48 hours while stirring and centrifuged. The supernatant liquid is recovered, mixed with cold acetone having 45% final concentration, the produced precipitate is recovered by centrifuging, dissolved in a buffer solution, concentrated by an ultrafilter, the fraction is successively purified by molecular sieve chromatography and immobilized heparin column chromatography to give the objective protein having about 16,000 dalton molecular weight by SDS-polyacrylamide gel electrophoresis and activity of multiplying cartilage cell alone or in the presence of a fibroblast growth factor.</p> |
