Nucleotide anhydride preparation

Nucleotide anhydrides are prepd. by reacting an alkylammonium salt of nucleoside-51-monophosphate in dioxan, DMF or dimethylacetamide with diphenyl phosphorochloridate or tetraphenyl pyrophosphate in the presence of an alkylamine. The mixture is then treated with a soln. of an alkylammonium salt of an acid, weaker than diphenyl phosphate, in pyridine or a pyridine deriv. The anhydrides have a wide range of applications depending upon the nature of the nucleoside.
