Compounds of formula (I), and salts and prodrugs thereof wherein: R<1> is H, certain optionally substituted C1-6alkyl, or C3-7cycloalkyl; R<2> represents a group (a) wherein X is O, S or NR<8> where R<8> is H or C1-4alkyl; one of Z and Y is C=O and the other is O, S or NR<9>, where R<9> is H or C1-4alkyl; R<3> is C1-6alkyl, halo or NR<6>R<7>; R<4> is C3-10cycloalkyl; n is 0, 1, 2, or 3; are CCK and/or gastrin receptor antagonists. They and compositions thereof are useful in therapy.
