PURPOSE:To efficiently obtain new histamine oxidase with high specificity, also excellent in the oxidative degradativity by culture, in a medium, of histamine oxidase-productive bacteria belonging to Arthrobacter. CONSTITUTION:The objective histamine oxidase can be obtained by culture of histamine oxidase-productive bacteria belonging to Arthrobacter [e.g. Arthrobacter globiformis IGO-12137 (ATCC-8010)] in a histamine-contg. medium. This enzyme has the following physicochemical characteristics: (1) activity: oxidative degradation of histamine; (2) substrate specificity: capable of degrading histamine, but incapable of degrading benzylamine, phenylpropylamine, putrescine, cadaverine, etc.; (3) optimal and stable pH range: optimal pH: 8.0 (in a sodium bicine buffer solution), stable pH: 7.0 (in a potassium phosphate buffer solution); and (4) molecular weight: 73000 (determined by high-performance liquid chromatography using a TSK gel G3000SW column). |
