PURPOSE:To provide the subject polypeptide exhibiting a human type caldesmon activity having a calmodulin affinity and an actin affinity, hopeful, e.g. for therapy of cancer, capable of being utilized for diagnosis of cancer and useful as a chimera protein or a transplantation material. CONSTITUTION:All the RNAs are extracted from Hela cells of human type cells by guanidine-cesium chloride method and a poly(A)<+>RNA is isolated by oligo(dT)-cellulose column chromatography. A cDNA is synthesized by using the isolated poly(A)<+>RNA as a template according to Gubler & Hofmann method and a cDNA library is prepared by using the resultant cDNA according to the conventional method. The obtained cDNA library is inserted into a host colibacillus so as to form a plaque. Screening is carried out by a probe in which a chicken type caldesmon fragment is used so as to select a strain containing a gene coding human type caldesmon. The gene is recovered therefrom and a recombination gene is prepared so as to transform a host. The resultant transformat is cultured, thus affording the objective polypeptide represented by the formula and exhibiting human type caldesmon activity. |
