PURPOSE:To industrially and advantageously obtain the subject compound participating in the agglutination of erythrocyte and intercellular recognition, etc., by reacting N-acetylneuraminic lyase in the presence of N-acetylglucosamine and pyruvic acid under alkaline conditions. CONSTITUTION:N-Acetylglucosamine and pyruvic acid are dissolved in water and the pH of the resultant solution is regulated to 10.5 with a 1N aqueous solution of sodium hydroxide. N-Acetylneuraminic lyase is then added to the solution under the alkaline conditions and made to react at 30 deg.C for 48hr. The resultant reactional product is subsequently isolated and concentrated by ion exchange column chromatography, etc. An organic solvent is added to form a needlelike crystal of the N-acetylneuraminic acid according to a conventional method. The formed crystal is then separated by filtration and dried to mass- produce the objective N-acetylneuraminic acid which is the most universal substance of sialic acids and yearly increasing the demand at a low cost. |
