PURPOSE:To obtain the subject enzyme suitable for providing a glucose from a liquidized starch by culturing a bacterium of the genus Streptomyces in a culture medium, producing and accumulating an acid resistant and heat resistant pullulanase in the cultured mixture and collecting the pullulanase from the cultured mixture. CONSTITUTION:A bacterium of the genus Streptomyces having ability capable of producing an acid resistant and heat resistant pullulanase is cultured to produce and accumulate the pullulanase in a culture medium and the objective acid resistant and heat resistant pullulanase is collected from the cultured mixture. The objective pullulanase has properties capable of hydrolyzing alpha-1,6-glucoside bond of pullulan to produce maltotriose and hydrolyzing alpha-1,6-glucoside bond of amylopectin. |
