PURPOSE:To efficiently obtain the subject growth hormone by constructing a secretory plasmid for the human growth hormone in a promoter, etc., for expressing a neutral protease gene of Bacillus.amyloliquefaciens and culturing a strain containing the plasmid in a specific culture medium. CONSTITUTION:A transformant of Bacillus subtilis transformed with a human growth hormonal secretory plasmid in which a DNA fragment having a mature human growth hormonal gene of 20000 daltons molecular weight bound just after a region of a promoter participating in the expression of a neutral protease gene derived from Bacillus.amyloliquefaciens, a ribosome binding site and a secretory signal is bound to a vector DNA composed of a plasmid replicable with the Bacillus subtilis is cultured in a culture medium containing 0.1-1.0% (wt./vol.) maltose or a culture medium containing 0.1-5% (wt./vol.) malt extract and 0.05-5% (wt./vol.) potassium citrate. Thereby, the objective human growth hormone of 20000 daltons molecular weight is secreted and produced in the culture supernatant. |
