DNA CODING PHOSPHOLIPASE A2, AND METHOD FOR PRODUCING PHOSPHOLIPASE A2 AND ITS PURIFICATION

PURPOSE:To massively obtain the phospholipase A2 in a short time in high purity and in high yield by culturing an insect cell transformed with a vector containing a specific DNA. CONSTITUTION:(A) A DNA coding a phospholipase A2 (PLA2) represented by the sequence of the formula is designed and synthesized so as to use a codon much used in insect cells as much as possible is obtained. An insect cell host such as a s.f.9 cell is transformed with a vector obtained by introducing the component A into a transfer vector such as pAc373 to obtain (B) a transformed insect cell. The cell B is infected with a recombinant virus and cultured in a medium to obtain (C) a crude PLA2 containing the PLA2. The component C is subjected to an affinity chromatography and to a reverse phase high performance liquid chromatography to provide the purified phospholipase A2.
