Cyclosporins wherein the residue at the 1-position has the formula I in which -x-y- is -CH=CH-(trans) or -CH2-CH2-, X is 0 or S and R is hydrogen, optionally halo or hydroxy substituted-C1-3-alkyl, -alkoxy or -alkylthio, or amino or mono- or di-(C1-2alkyl)-amino, useful as pharmaceuticals, preferentially for topical administration in particular for asthma therapy.
